Ham's F-12K medium was originally designed to grow primary human hepatocytes as well as differentiated rat and chicken cells.

Phenol red is used as an indicator of the pH value in the culture medium to continuously monitor the pH of the culture solution. At low pH, phenol red makes the culture solution yellow, and at higher pH values it makes the culture solution purple. , It is red when the pH value is 7.2 to 7.4, which is most suitable for cell culture. But phenol red also has some disadvantages. Studies have shown that phenol red can mimic the effect of steroid hormones (especially estrogen). Therefore, when using estrogen-sensitive cells (such as breast tissue), it is best to use phenol red-free Medium. Phenol red will interfere with the detection during flow cytometry. In addition, the presence of phenol red in the formulation of some serum-free media can interfere with the sodium-potassium balance.
